Fluorescent Agonists of the alpha7 Nicotinic Acetylcholine Receptor Derived from 3-Amino-Quinuclidine

Here, we investigated whether fluorescence labeled small molecule agonists of the alpha7 nicotinic acetylcholine receptor (nAChR) might be identified to enhance receptor studies. Enantiomerically pure 3-amino-quinuclidines appended with fluorophores at the 3-amino group were synthesized and tested by electrophysiology on human alpha7 nAChR in Xenopus oocytes, uncovering (R)-4 and (R)-9 as the first examples of fluorescent alpha7 nAChR agonists. These molecules elegantly incorporate the fluorescent reporter group as part of the pharmacophore itself and provide a new class of tool compounds for the study of these ligand-gated ion channels.

Directed synthesis of noncentrosymmetric molybdates using composition space analysis

A systematic investigation of the factors governing the reaction product composition, hydrogen bonding, and symmetry was conducted in the MoO 3/3-aminoquinuclidine/H2O system. Composition space analysis was performed through 36 individual reactions under mild hydrothermal conditions using racemic 3-aminoquinuclidine. Single crystals of three new compounds, [C7H16N2][Mo 3O10]¡¤H2O, [C7H 16N2]2[Mo8O26] ¡¤H2O, and [C7H16N2] 2[Mo8O26]¡¤4H2O, were grown. The relative phase stabilities for these products are dependent upon the reactant mole fractions in the initial reaction gel. This phase stability information was used to direct the synthesis of two new noncentrosymmetric compounds, using either (S)-(-)-3-aminoquinuclidine dihydrochloride or (R)-(+)-3-aminoquinuclidine dihydrochloride. [(R)-C7H 16N2]2-[Mo8O26] and [(S)-C7H16N2]2[Mo8O 26] both crystallize in the noncentrosymmetric space group P2 1 (No. 4), which has the polar crystal class 2 (C2). The second-harmonic generation activities were measured on sieved powders. The structure-directing properties of the molybdate components in each compound were determined using bond valence sums. The structures of all five compounds were determined using single-crystal X-ray diffraction.

Discovery and structure-activity relationship of quinuclidine benzamides as agonists of alpha7 nicotinic acetylcholine receptors

A library of benzamides was tested for alpha7 nicotinic acetylcholine receptor (nAChR) agonist activity using a chimeric receptor in a functional, cell-based, high-throughput assay. From this library, quinuclidine benzamides were found to have alpha7 nAChR agonist activity. The SAR diverged from the activity of this compound class verses the 5-HT3 receptor, a structural homologue of the alpha7 nAChR. PNU-282987, the most potent compound from this series, was also shown to open native alpha7 nAChRs in cultured rat neurons and to reverse an amphetamine-induced gating deficit in rats.

Synthesis and biological activities of indolizine derivatives as alpha-7 nAChR agonists

Human alpha7 nicotinic acetylcholine receptor (nAChR) is a promising therapeutic target for the treatment of schizophrenia accompanied with cognitive impairment. Herein, we report the synthesis and agonistic activities of a series of indolizine derivatives targeting to alpha7 nAChR. The results show that all synthesized compounds have affinity to alpha7 nAChR and some give strong agonistic activity, particularly most active agonists show higher potency than control EVP-6124. The docking and structure-activity relationship studies provide insights to develop more potent novel alpha7 nAChR agonists.
